Since the Update of iTunes 12, the function is for the random Genius list is a little hidden:

Start iTunes and check if Genius is activated. To do this, go in the menu bar, click "Store" - > "Genius enable/disable".
Keep Windows [Ctrl]-key and under MacOS, the [alt] key pressed. Now, the Genius icon appears at the point of rewind/Last track Icon. Click on this to generate a random Genius list.
The created Playlist you can view with a click on the Playlist icon (three horizontal bars with dots in front of it). Here, you can create with a click on "Re-happen again," a new random Genius list.
